主要科研领域与方向

1. 冷冻电镜断层成像(cryo-ET)及原位结构生物学。高分辨cryo-ET是通过收集三维断层图像,再使用子断层图像平均法进行结构解析的前沿结构生物学方法。 Cryo-ET擅长对柔性巨型分子机器、细菌、细胞进行成像和蛋白结构解析,获得具生理意义的原位结构,是结构生物学的下一个突破。

2. 新发致病型囊膜病毒的组装及通过膜融合解胞的结构与功能。由于形态不均一,囊膜病毒的原位结构一直是结构生物学的挑战。我们使用cryo-ET解析全病毒结构,及在细胞中直接观测病毒生命活动的原位结构,希望回答:囊膜病毒如何组装,又如何通过膜融合解胞?目的找到病毒的弱点,并为抗体、疫苗设计提供新思路。
